Inhibition of Neuroinflammation in LPS-Activated Microglia by Cryptolepine

Cryptolepine (CAS) inhibited PGE2 release and COX-2 protein expressions in LPS-activated microglia. Microglia were incubated in a medium containing 2.5 and 5 μM cryptolepine for 30 min and then activated by 100 ng/mL LPS for 24 h. (a) Cryptolepine diminished PGE2 release in microglia. ((b), (c)) Cryptolepine inhibited COX-2 but not mPGES1 protein expression in LPS-activated microglia. Protein expression was determined using western blot with specific anti-COX-2 and anti-mPGES-1 antibodies. All values are expressed as mean ± SEM for 3 independent experiments. Data were analysed using one-way ANOVA for multiple comparisons with post-hoc Student Newman-Keuls test. * in comparison with LPS control.
