(a) PC12 cells were pretreated with various concentrations of D for 3 h and then incubated with and without 25 μM Aβ for 24 h. Cell viability was estimated by the WST-8 assay. Effect of D on Aβ -induced intracellular ROS generation in PC12 cells. (b) PC12 cells were pretreated with various concentrations of D for 3 h and then incubated with and without 25 μM Aβ for 24 h. After that the cells were washed with PBS and then treated with 10 μM H2DCF-DA for 30 min. (c) PC12 cells were incubated with D (10μM) for the indicated time. They were then treated with 25 μM Aβ for 24 h and with 10 μM H2DCF-DA for 30 min. Intracellular ROS production was measured at the excitation wavelength of 485 nm and the emission wavelength of 530 nm using the fluorescence microplate reader.
