The Effects of Lipid Membranes, Crowding and Osmolytes on the Aggregation, and Fibrillation Propensity of Human IAPP

Figure 6

Visual leakage assay studying the interaction of hIAPP with biological membrane and its consequence. Confocal fluorescence microscopy images of the interaction of 5 μM hIAPP with giant unilamellar vesicles (GUVs) composed of lipids extracted from a pancreatic β-cell line of rat (INS-1E). The GUVs which are labelled with N-Rh-DHPE (red) are filled with phosphate buffer containing the fluorophore Atto647 (blue). C-terminally labelled hIAPP-K-Bodipy-FL (green) adsorbs within the first 5 min to the lipid vesicles and leads to membrane permeabilization and disintegration of the GUV. The scale bars represent 10 μm. Reprinted from [60] with permission from Elsevier.
