Contractile activity is essential for iNOS expression. (a) NRVMs were treated with L-type calcium channel blockers (nifedipine 5–10 μM or verapamil 5–10 μM) or myosin II ATPase inhibitors (butanedione monoxime (BDM) 3.75–7.5 mM or blebbistatin 5–10 μM) for 24 h. Cell extracts (50 μg total protein per lane) were then separated by SDS-PAGE and Western blotting with antibodies specific for iNOS, eNOS, and GAPDH.
