In normoxia, HIF1α is hydroxylated by PHD. Hydroxylated HIF1α undergoes protein degradation via vHL proteasome pathway. In hypoxia, PHD does not hydroxylate HIF1α. Stabilized HIF1α is translocated across nucleus membrane dimerizing with HIF1β. Dimerization of HIF1α/β increases HIF1 transcription factor which binds to HRE of DNA.
